一、个人简介
科研成果
齐以涛,博士,博士生导师,陕西省百人计划特聘教授。2007年7月毕业于中国农业大学获得博士学位。2007年8月至2012年7月在美国德州心脏研究所(Texas Heart Institute)从事博士后研究,2017年8月至2014年12月在美国德州心脏研究所任Research associate。2015年1月至今任买球平台官方网站教授,主要研究蛋白质的SUMO修饰参与发育和疾病发生发展的分子机制,相关研究成果以第一或通讯作者身份在Mol Cell、Neuron、Cell Death Differ、Int J Biol Macromol和J Biol Chem等杂志发表研究论文50余篇,并获得美国MD Anderson Cancer Center“杰出论文奖”。研究成果得到了许多国际主流媒体的关注和报道,包括ScienceDaily和EurekAlert等。已申请发明专利1项,发表学术著作1部,获得多项国家和陕西省自然科学基金项目的资助。

二、主要研究领域及兴趣
SUMO修饰(SUMOylation)是一种和泛素化修饰类似的蛋白质修饰形式,影响其所修饰的蛋白质功能与定位的一个关键性调节机制。它参与调节多种细胞生命活动过程,包括细胞周期、细胞分裂、DNA损伤修复、细胞内转运、调节蛋白稳定和基因转录等。SUMO修饰是一个动态可逆的过程,由SUMO特异的活化酶(E1)、结合酶(E2)和连接酶(E3)相继作用来催化完成,其去SUMO修饰的过程(De-SUMOylation)则是由SUMO特异性蛋白酶家族(SENP)来完成。本实验室主要研究蛋白质的SUMO修饰, 特别是由SENP2介导的去SUMO修饰过程和对细胞信号转导的调控机制,以及它们在发育与疾病过程中的作用与意义。主要研究领域有:
1. SENP2的靶蛋白分子与生物学功能
SENP2的功能是通过对靶蛋白质的去SUMO化,来改变靶蛋白的活性来实现的。因此,研究SENP2的生物学功能,首先要鉴定SENP2的靶蛋白分子。我们通过功能性筛选和基因敲除等策略来寻找和鉴定SENP2的靶蛋白质,并研究SENP2调控靶蛋白的作用与机制,从而来明确SENP2的生物学功能。
2. 调控SENP2的细胞信号网络
SENP2的去SUMO化参与了许多细胞生命活性过程,也会受到细胞信号网络的调控。因此我们在鉴定SENP2靶蛋白的基础上,研究细胞信号通路对这一去SUMO化过程的调节作用与机制,并依此建立SENP2的细胞信号调控网络。
3. SENP2在发育与疾病中的作用与意义
SUMO修饰的靶蛋白分子与发育和疾病有密切的关系,因此SENP2通过这些靶蛋白分子的去SUMO化修饰,必将参与对发育和疾病的调控。我们利用细胞、模式动物及疾病标本,综合运用分子、细胞和发育生物学以及病理学的知识与手段,研究蛋白质的SUMO修饰在发育与疾病过程中的作用机制,为医学实践提供理论基础。
